You have to check is there any damages in SPen. Hope you already did "reset S pen" as shown in the attached screenshot. If not try once. Even it's not connected then its SPen fault. For me it happened once on another note.. A toggle to airplane mode n back fixed it..

How to Reset S Pen
Please follow the below steps:

Step1: Navigate to the Apps screen and tap on Settings.
Step2: Tap on Advanced features.
Step3: Tap on S Pen.
Step4: Tap on Air actions.
Step5: Tap on More.
Step6: Tap on Reset S Pen.

In case the issue still occurs. Please register your concern in Samsung Members Application (Open Samsung Members Application > Get help > Send Feedback > Error report/Ask questions).
