Can I lock any specific application without using 3rd party apps.?

Samsung's S Secure from Galaxy Story. Once installed, you can use it from Settings > Advanced Features

Unfortunately not. There used to be an app from Samsung for this, called S Secure, but they discontinued it for most phones...
