Hi... try these steps.
First go to Get Help tab on this app and select Diagnostic. Then select USB connection. Run the test and check whether it's working normally. (As in the Screenshot)
If you dont get message like in the Screenshot, Try to use a different OTG adapter and a pen drive.
Still you have the issue, rest to your device. But still it's not working, it could be a hardware issue. Then you have to contact a Samsung service center near to you.

This happened to me once as well then what I did was I format the pen drive before pluging it via OTG. Then what I did was I enabled USB debugging. Follow these screenshot. To enable it you need to tap on Build number 4 to 5 times then it will enable
Go to Setting > About > Software information > Build number
