There will be a pop up in your computer screen, there you can select Open device to view files. You can use it. For OTG there is no other setting. u can connect and start using. if it is still not working then try that pendrive in any other phone compatible for OTG. if it works there then it should work here. still if it is not working then get the port checked at service center. they will help you.
