Yes there are major differences between bth the UI.

One UI core doesn't get the KNOX SECURITY features on all devices ... It doesn't get the Altz life, Secure folder, Hardware grade security....
.

Well, simply put, you won't be getting all One UI features in One UI Core. There are no compromises in terms of security. It all comes down to what you want. Optimization is better in One UI Core because it is lighter on RAM and processor.

One major difference is that you cannot install good lock or most of its modules on One UI core, while on One UI you can sideload it if not available in galaxy store.
Most of the S series get all the new features that samsung releases, while A series may or may not get it later. But they don't cut down most features a normal person uses. S series are like luxury phones, while A series have all the necessary features, and most M series trade One UI full version with core for better camera, battery, etc.

You cannot update samsung apps manually on OneUI core... because it uses old version of those apps.
Samsung hides all its customisation behind fancy name 'goodlock' whose many features are given by chinese by default.
It have less features than stock android,
It is so bad that, newer M & F models are having normal version.
